Overview

Ad5-Brachyury (ETBX-011) is a therapeutic cancer vaccine developed by ImmunityBio, utilizing a modified, non-replicating adenovirus type 5 (Ad5) vector to deliver the gene for the human Brachyury protein. Brachyury is a transcription factor and a key driver of the epithelial-mesenchymal transition (EMT), which is nearly universally expressed in chordoma and frequently overexpressed in various other solid tumors. The vaccine is designed to stimulate the immune system, specifically inducing a Brachyury-specific T-cell response to identify and destroy tumor cells. In the context of chordoma, it is often evaluated as part of a complex multi-modal immunotherapy regimen (the NANT regimen) that includes other vaccines, cytokines like Anktiva (N-803), and natural killer cell therapies.
