Ad5-CEA-NIS is an experimental gene therapy vector developed by the University of Würzburg for the treatment of carcinoembryonic antigen (CEA)-expressing cancers, specifically medullary thyroid cancer. It utilizes a replication-deficient adenovirus serotype 5 (Ad5) to deliver the human sodium-iodide symporter (hNIS) gene. The expression of hNIS is transcriptionally targeted to tumor cells using the CEA promoter. Once expressed on the surface of tumor cells, hNIS facilitates the uptake of radioisotopes such as iodine-131 (¹³¹I), enabling targeted internal radiotherapy in tumors that do not naturally concentrate iodine. This approach aims to sensitize medullary thyroid cancer cells, which are typically refractory to standard radioiodine therapy, to radionuclide treatment.
