Overview

The Ad5-CEA vaccine is a therapeutic cancer vaccine that utilizes a modified adenovirus serotype 5 (Ad5) vector. This vector is engineered with deletions in the early 1 (E1), early 2b (E2b), and early 3 (E3) viral genes, which helps to reduce the host's immune response to the vector itself and allows for robust expression of the transgene. The vaccine delivers the gene for carcinoembryonic antigen (CEA), a tumor-associated antigen (TAA) that is frequently overexpressed in various cancers, including colorectal, lung, breast, and pancreas cancers. Upon administration, the expressed CEA protein aims to induce a potent cytotoxic T-lymphocyte (CTL)-mediated immune response specifically against tumor cells that express CEA, thereby inhibiting tumor cell proliferation and promoting tumor cell death. Developed by ImmunityBio (through its subsidiary Etubics) in collaboration with the National Cancer Institute and National Institutes of Health, Ad5-CEA vaccine has been investigated both as a standalone agent and as a component of multi-antigen vaccine regimens, such as the Tri-Ad5 vaccine, which also targets MUC1 and brachyury.
