Ad5-CMV-mTNFa is a replication-deficient adenoviral vector (serotype 5) designed to deliver the murine tumor necrosis factor-alpha (mTNF-α) gene under the control of the constitutive cytomegalovirus (CMV) promoter. TNF-α is a potent cytokine that induces apoptosis in cancer cells, activates the vascular endothelium, and stimulates the immune system. By utilizing an adenoviral vector, researchers can achieve high levels of localized TNF-α expression within the tumor microenvironment, which aims to maximize anti-tumor efficacy while minimizing the systemic toxicity typically associated with recombinant TNF-α protein administration. This construct is primarily used as a research tool in preclinical oncology to study the effects of localized cytokine therapy and its synergy with other modalities such as radiotherapy or chemotherapy in syngeneic mouse models.
