Ad5-CMV-NIS is a recombinant, replication-deficient adenovirus serotype 5 (Ad5) vector engineered to express the human sodium-iodide symporter (NIS) gene under the control of the cytomegalovirus (CMV) promoter. The NIS protein enables cells to actively uptake iodide, which can be exploited for targeted radiovirotherapy using radioactive iodine isotopes such as I-131 or I-123. This approach allows for selective imaging and ablation of transduced tumor cells following viral delivery. The primary mechanism involves gene transfer-mediated expression of NIS in cancer cells, facilitating their destruction by subsequent administration of therapeutic radioisotopes. Preclinical studies have demonstrated efficacy in various tumor models including prostate and liver cancers[1][3][4][9]. The construct is primarily used as an investigational gene therapy tool for cancer treatment and imaging.
