**Ad5-D24-PEPvIII** is a preclinical, EGFRvIII-directed oncolytic adenovirus-based cancer vaccine developed for malignant glioma, including glioblastoma. The construct displays multiple copies of the epidermal growth factor receptor variant III peptide antigen PEPvIII in the adenoviral hexon protein, coupling local oncolytic-virus activity with vaccination against EGFRvIII-positive tumor cells. In immunocompetent mouse glioma models, local administration induced PEPvIII-specific T-cell responses; enhanced derivative constructs expressing GM-CSF and soluble PD-1, as well as combination with PD-L1 blockade, were investigated separately.
