Overview

Ad5-D24RGD is a conditionally replicative oncolytic adenovirus (CRAD) derived from adenovirus serotype 5, engineered for enhanced tumor selectivity and infectivity. The "D24" (or Delta-24) modification consists of a 24-base pair deletion in the E1A gene, which prevents the virus from binding to the Retinoblastoma (Rb) protein. This restriction ensures that viral replication occurs selectively in cancer cells with a dysfunctional Rb-p16 pathway, while sparing normal cells. To overcome the low expression of the primary Coxsackie-Adenovirus Receptor (CAR) in many advanced tumors, the virus features an "RGD" modification—an RGD-4C peptide motif inserted into the HI-loop of the fiber knob. This allows the virus to utilize $\alpha_v\beta_3$ and $\alpha_v\beta_5$ integrins as alternative receptors for cell entry. Developed primarily by researchers at the University of Helsinki, Ad5-D24RGD has been investigated for the treatment of various solid tumors, including cervical and ovarian cancers, and serves as the scientific basis for clinical-stage agents like DNX-2401.
