Ad5-delta24RGD is a conditionally replicative oncolytic adenovirus (CRAd) developed by the University of Alabama at Birmingham for the treatment of gynecologic malignancies, specifically recurrent ovarian and primary peritoneal cancers. The virus is engineered with two primary modifications to enhance its therapeutic index: a 24-base pair deletion in the E1A gene (Δ24), which restricts viral replication to cancer cells with a dysfunctional Retinoblastoma (Rb)/p16 pathway, and the incorporation of an RGD-4C motif into the fiber HI loop. This RGD modification allows the virus to infect cells via αvβ3 and αvβ5 integrins, which are frequently overexpressed in ovarian cancer, thereby bypassing the need for the Coxsackie adenovirus receptor (CAR). Administered intraperitoneally, Ad5-delta24RGD aims to selectively replicate within and lyse tumor cells while sparing healthy tissue.
