Overview

Ad5-hSulf1 is a recombinant adenovirus serotype 5 (Ad5) vector engineered to express the human **sulfatase 1 gene (hSulf-1)**. The drug is designed for cancer gene therapy; its mechanism of action centers on the **restoration of hSulf-1 function** in tumor cells, which is often downregulated in cancers. By expressing hSulf-1, Ad5-hSulf1 negatively regulates cell signaling pathways downstream of growth factors by desulfating heparan sulfate proteoglycans, thereby reducing growth factor signaling such as AKT and ERK phosphorylation, downregulating proliferative and anti-apoptotic molecules (Cyclin D1, Survivin), and resulting in cell cycle arrest, increased apoptosis, and inhibition of angiogenesis. It has been shown to suppress tumor growth in preclinical models of hepatocellular carcinoma, melanoma, ovarian, and other cancers[2][4][6].
