Ad5-IFNA4 is an experimental gene therapy consisting of a replication-deficient recombinant adenovirus type 5 (Ad5) vector that carries the human interferon-alpha 4 (IFNA4) gene. This modality is designed to deliver the IFNA4 gene directly into target cells, such as the bladder epithelium or respiratory mucosa, where it induces the local production and secretion of the interferon-alpha 4 cytokine. Interferon-alpha 4 is a member of the Type I interferon family and possesses potent anti-tumor, anti-proliferative, and immunomodulatory activities. By utilizing a viral vector for localized delivery, the therapy aims to achieve high therapeutic concentrations of the cytokine at the site of disease while minimizing the systemic toxicity typically associated with recombinant interferon protein administration. Ad5-IFNA4 has been primarily investigated in preclinical and early-stage clinical research for the treatment of non-muscle invasive bladder cancer (NMIBC) and chronic hepatitis B (CHB), as well as for prophylaxis against respiratory viral infections like COVID-19.
