Ad5-MuLV gp70 is a preclinical recombinant adenovirus serotype 5 (Ad5) viral vector vaccine encoding the envelope glycoprotein gp70 of the murine leukemia virus (MuLV). Developed by researchers at the University Hospital Erlangen, this vaccine is designed to target endogenous retroviral antigens (such as MelARV gp70) that are aberrantly expressed in various murine cancer models, including the 4T1 triple-negative breast cancer model. When administered mucosally (intranasally), Ad5-MuLV gp70, often co-delivered with an Interleukin-1β-encoding vector or other tumor-associated antigen vectors like Ad5-Mage-b, robustly induces tumor-specific tissue-resident memory T cells (TRM) in the lungs. This localized immune response has been shown to significantly prevent and treat pulmonary metastases and improve survival in preclinical breast cancer models.
