Ad5-PC is a novel, replication-competent oncolytic adenovirus (serotype 5) engineered with an E1B 55-kDa deletion to selectively replicate in p53-mutated cancer cells, leading to tumor cell lysis. It serves as a gene therapy platform that expresses a bispecific soluble fusion protein containing the extracellular domains of Programmed cell death protein 1 (PD-1) and CD137 ligand (CD137L). This fusion protein is designed to overcome tumor-induced immunosuppression by simultaneously blocking the PD-1/PD-L1 checkpoint and providing costimulatory signals through the CD137 receptor. Preclinical studies in hepatocellular carcinoma models demonstrate that Ad5-PC increases the infiltration and persistence of activated CD8+ T cells and interferon-gamma production within the tumor microenvironment, resulting in significant tumor regression and systemic antitumor immunity.
