Ad5.F35-GUCY2C-PADRE is a recombinant cancer vaccine designed to prevent recurrence in patients with gastrointestinal (GI) adenocarcinomas following definitive surgery and adjuvant therapies. The vaccine uses a chimeric viral vector composed of an adenovirus serotype 5 (Ad5) capsid and an adenovirus serotype 35 (Ad35) fiber protein, which reduces sensitivity to pre-existing immunity against Ad5. It encodes the extracellular domain of guanylyl cyclase C (GUCY2C), a protein overexpressed in GI cancers but compartmentalized in normal intestinal cells, making it an attractive immunotherapy target. The construct also includes PADRE, a universal CD4+ T-helper epitope that enhances immune response. Upon intramuscular administration, the vector targets CD46 on tumor cells for uptake and induces anti-tumor immunity by presenting GUCY2C antigens to the immune system[1][3][4][8].
