AD55-Mn-SOD is a **dual-regulated oncolytic adenovirus** constructed to deliver the manganese superoxide dismutase (Mn-SOD) gene for cancer therapy, specifically targeting hepatocellular carcinoma. This agent employs a liver cancer-specific α-fetoprotein (AFP) promoter to drive tumor-selective replication of the adenovirus, which enables the virus to deliver and overexpress Mn-SOD in hepatocellular carcinoma (HCC) cells. Mn-SOD acts as a tumor suppressor by promoting oxidative stress-mediated apoptosis in cancer cells, with the AD55-Mn-SOD vector specifically inducing cell death via the activation of the caspase apoptotic pathway. In preclinical models, AD55-Mn-SOD demonstrated tumor-specific cytotoxicity and suppressed the growth of HCC xenografts in vivo[1]. This approach represents a gene-virotherapy strategy, integrating mechanisms of viral oncolysis with targeted gene delivery for enhanced anticancer activity.
