Ad5Ag85A is an intramuscularly injected recombinant replication-deficient adenovirus vector (Ad5)-based vaccine that expresses a Mycobacterium tuberculosis antigen Ag85A. It is designed as a heterologous booster vaccine following BCG priming. In animal models, Ad5Ag85A showed markedly improved protection over BCG alone, with respiratory route administration providing the best type of protection. In human clinical trials, intramuscular vaccination was found to be safe, immunogenic, and capable of stimulating polyfunctional T cell responses, more potently in previously BCG-vaccinated volunteers. Pre-existing Ad5 antibodies did not significantly dampen the immune response.
