Ad5CMV-p53 + cisplatin is a combination therapy consisting of an adenoviral vector containing the wild-type p53 gene (Ad5CMV-p53) and the traditional chemotherapy drug cisplatin. This combination works synergistically to induce apoptosis in cancer cells. Ad5CMV-p53 introduces functional p53 protein into cancer cells, which can restore normal p53 tumor suppressor activity, while cisplatin acts as a DNA-damaging agent. The combination has shown greater cytotoxicity (approximately 25% more) compared to cisplatin alone in various cancer cell lines, including those with mutant p53. The synergistic effect appears to be due to p53-mediated sensitization of cancer cells to cisplatin-induced cell death.
