Overview

AD5CON is a **replication-competent recombinant oncolytic adenovirus type 5** used in preclinical studies as a control or benchmark oncolytic virus. It acts as a non-transgene-expressing comparator to engineered adenoviruses carrying therapeutic transgenes, for instance, in studies involving adenoviruses expressing immunostimulatory proteins such as soluble PD-1, CD137L, or PVR. AD5CON induces moderate immune responses in the tumor microenvironment, increasing infiltration of immune cells like CD8+ T cells and NK cells, and can convert "cold" tumors (immunologically inactive) into more "hot" (immunologically active) ones. However, in vivo studies show that AD5CON alone is generally **insufficient to produce substantial antitumor effects**, likely due to upregulation of inhibitory signals (e.g., PD-L1) on tumor cells. Therefore, it is mainly used as a negative control in adenoviral immunotherapy research, rather than as a candidate for clinical development[1][3].
