Ad5D24-CpG is a genetically engineered oncolytic adenovirus based on the human adenovirus serotype 5 backbone, featuring a 24-base pair deletion in the E1A region (D24) and multiple synthetic CpG motifs inserted into its genome. The inclusion of CpG islands increases the virus’s capacity to stimulate the immune system via Toll-like receptor 9 (TLR9), enhancing both innate and adaptive antitumor immune responses. As an oncolytic virus, Ad5D24-CpG specifically infects and lyses tumor cells, releasing tumor antigens and stimulating an antitumor immune response. Preclinical studies indicate increased NFκB activation, robust NK and T cell-mediated tumor cell killing, and improved tumor control compared with conventional oncolytic adenoviruses. It is under development primarily for cancer immunotherapy applications[2][3].
