Ad5f35pp65-stimulated donor-derived cytotoxic T lymphocytes is an investigational adoptive cellular immunotherapy developed to prevent cytomegalovirus (CMV) reactivation in patients following allogeneic hematopoietic stem cell transplantation (HSCT). The therapy involves the ex vivo generation of CMV-specific cytotoxic T lymphocytes (CTLs) from donor peripheral blood. This is achieved by stimulating donor cells with dendritic cells that have been transduced with an adenoviral vector (Ad5f35pp65) encoding the CMV pp65 antigen. The resulting CTLs are then infused into the patient to reconstitute cellular immunity against CMV, targeting and eliminating cells expressing the pp65 protein.
