Ad5sPVR is a **recombinant oncolytic adenovirus** engineered to express a soluble extracellular domain of the poliovirus receptor (sPVR). Developed as a novel oncolytic virotherapy, Ad5sPVR selectively replicates in and lyses tumor cells while simultaneously expressing sPVR to modulate local immune responses. The soluble PVR protein binds the inhibitory checkpoint receptor TIGIT with high affinity, blocking the PVR/TIGIT pathway and relieving T cell inhibition. It also activates the activating receptor CD226, thereby enhancing antitumor immune responses. In preclinical models (including H22 hepatocellular carcinoma ascites and B16/F10 solid tumors), Ad5sPVR increased CD8+ T cell and NK cell infiltration, promoted interferon-gamma release, improved immune surveillance, and showed a durable antitumor effect. The drug is considered a potential next-generation oncolytic virus with immune checkpoint blockade properties, primarily studied for cancer immunotherapy, notably in hepatocellular carcinoma and melanoma models[1].
