Ad5VLP is a next-generation adenovirus-derived virus-like particle (VLP) platform designed for the rapid delivery of antigenic mRNA or protein cargo. Developed by researchers at Washington University in Saint Louis, the system utilizes a helper-virus-based approach where the adenoviral core protein pV is fused to the RNA-binding protein MCP to facilitate the encapsulation of foreign RNA, such as linear or circular mRNA. The platform combines the immunostimulatory properties of adenoviral structural proteins with the speed and scalability of VLP manufacturing, avoiding the lengthy rescue and amplification steps required for conventional recombinant adenoviral vectors. Preclinical studies have demonstrated its ability to induce robust antigen-specific humoral (B-cell) and cellular (T-cell) immune responses, making it a versatile candidate for infectious disease outbreak response and personalized cancer neoantigen vaccines.
