Drug intelligence / Profile preview
Ad825 is a novel tumor microenvironment (TME) gene-engineering adenoviral vector developed by Lokon Pharma (also referred to as LOK Enterprises). Designed as an immunostimulatory gene therapy, Ad825 is engineered to express three distinct transgenes directly within the TME: a trimerized membrane-bound CD40 ligand (TMZ-CD40L), a 4-1BB ligand (4-1BBL), and a secreted PD-L1-blocking Affibody molecule. This multi-pronged approach aims to activate CD40 on antigen-presenting cells, stimulate 4-1BB on T cells and natural killer (NK) cells, and block the immunosuppressive PD-1/PD-L1 checkpoint axis. Ad825 is currently in preclinical development, with research demonstrating its potential to drive anti-tumor immune responses and reduce factors promoting tumor progression in soft-tissue sarcoma.
