ADA-409 is a proprietary small molecule developed by Aranda Pharma that acts as a potent inhibitor of lipid peroxidation and protects against ferroptotic cell death. It is an arylthiazine (benzo[b]thiazine) derivative designed to provide neuroprotection in vitro and in vivo. The compound has demonstrated efficacy in inhibiting tert-butyl hydroperoxide-induced lipid peroxidation and protecting neuronal cells from ferroptosis triggered by glutathione depletion or glutathione peroxidase 4 (GPx4) inhibition. Additionally, ADA-409 suppresses pro-inflammatory activation of microglia and reduces infarct volume, edema, and expression of pro-inflammatory genes in animal models of thromboembolic stroke. Its primary therapeutic focus is on brain diseases where ferroptosis plays a key role—such as Parkinson’s disease, Alzheimer’s disease, ALS (amyotrophic lateral sclerosis), acute brain insults (e.g., stroke), neurodegenerative disorders, peripheral ischemia, and potentially prostate cancer[1][2][3][4].
