Overview

Adakitug is a fully human IgG1 kappa monoclonal antibody that binds with high affinity to the pro‑inflammatory chemokine interleukin‑8 (IL‑8, also known as CXCL8), neutralizing its interaction with IL‑8 receptors and thereby inhibiting IL‑8–driven neutrophil recruitment, angiogenesis, and tumor‑promoting inflammation.[1][4][7][9][11][15] Originally developed by Medarex and later advanced by Bristol Myers Squibb, the antibody (also referred to as BMS‑986253 or MDX‑018) has been investigated mainly as an antineoplastic and immuno‑inflammatory agent in Phase 1–2 trials for advanced solid tumors, hormone‑sensitive and castration‑sensitive prostate cancer, non‑small cell lung cancer, hepatocellular carcinoma, head and neck squamous cell carcinoma, melanoma, and COVID‑19, often in combination with immune checkpoint inhibitors such as nivolumab and ipilimumab.[1][4][6][12][18] By blocking IL‑8 signaling, adakitug is intended to reduce tumor cell mesenchymal features, dampen immunosuppressive myeloid infiltration, and modulate inflammatory pathways, with additional exploratory development in autoimmune and inflammatory diseases supported by its IL‑8–neutralizing profile.[1][2][7][9][15]
