Overview

This is a combination of three monoclonal antibody drugs—adalimumab, golimumab, and infliximab—all of which are anti-tumor necrosis factor alpha (anti-TNFα) agents. Each drug individually binds to and neutralizes tumor necrosis factor alpha (TNFα), a pro-inflammatory cytokine involved in the pathogenesis of several autoimmune diseases such as rheumatoid arthritis, Crohn's disease, ulcerative colitis, ankylosing spondylitis, and psoriatic arthritis[1][3][7]. These agents reduce inflammation by blocking TNFα-mediated signaling pathways that drive immune cell activation and inflammatory cytokine production. The combination of these three specific anti-TNF monoclonal antibodies is not an established or approved therapeutic regimen; there are no clinical guidelines or evidence supporting their concurrent use together due to overlapping mechanisms and increased risk for immunosuppression[4]. Each agent has been developed independently for similar indications but is typically used as monotherapy or in combination with non-biologic immunosuppressants.
