Overview

This is a combination of three distinct tumor necrosis factor (TNF) inhibitors—adalimumab, etanercept, and infliximab. Each is a biologic agent used individually to treat autoimmune and inflammatory diseases such as rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis, Crohn's disease (infliximab and adalimumab), ulcerative colitis (infliximab and adalimumab), plaque psoriasis, juvenile idiopathic arthritis, and others. All three drugs act by binding to TNF-alpha—a pro-inflammatory cytokine—and blocking its interaction with TNF receptors on cell surfaces. This inhibition reduces inflammation and alters immune responses associated with chronic autoimmune conditions[1][2][3][4][6][7]. - Adalimumab is a fully human monoclonal antibody administered subcutaneously[3]. - Etanercept is a fusion protein consisting of the extracellular ligand-binding portion of the human TNF receptor linked to the Fc portion of human IgG1; it is also given subcutaneously[4]. - Infliximab is a chimeric monoclonal antibody administered intravenously[7]. There are no approved pharmaceutical products that combine all three agents into one formulation or regimen; their concurrent use together would be highly unusual due to overlapping mechanisms and increased risk for immunosuppression.
