ADAM9-ADC refers to a class of antibody-drug conjugates (ADCs) designed to target ADAM metallopeptidase domain 9 (ADAM9), a transmembrane protein overexpressed in various solid tumors, including pancreatic, non-small cell lung (NSCLC), gastric, and colorectal cancers. ADAM9 mediates the proteolytic shedding of cytokines and growth factors, promoting tumor progression and metastasis. This class includes next-generation candidates, such as those developed by Henlius Biotech, which utilize camptothecin-based linker-payloads (topoisomerase I inhibitors). These newer agents are engineered to overcome the limitations of earlier ADAM9-targeted ADCs, such as IMGC-936, which was terminated due to maytansinoid-related ocular toxicities. By employing potent payloads with bystander killing effects and high plasma stability, ADAM9-ADCs aim to provide a superior therapeutic index for patients with ADAM9-positive malignancies.
