**ADAM9-MSN** is a preclinical, pancreatic ductal adenocarcinoma-targeted mesoporous silica nanoparticle drug-delivery platform. It uses an avidin-biotin gatekeeper system with an ADAM metallopeptidase domain 9-cleavable peptide linker, intended to release encapsulated chemotherapy preferentially in ADAM9-enriched pancreatic tumor microenvironments. Paclitaxel-loaded ADAM9-MSNs were cytotoxic to pancreatic cancer cells in vitro, but later preclinical studies found off-target toxicity and no meaningful in vivo antitumor response despite optimized formulations, prompting development of intracellular-protease-responsive alternatives.
