AdAPT-001 is an investigational oncolytic virus therapy based on a replication-competent type 5 adenovirus engineered to express a transforming growth factor beta (TGF-β) receptor-immunoglobulin fusion protein ("TGF-β trap"). This design enables the virus to selectively infect and lyse tumor cells while locally neutralizing the immunosuppressive and profibrotic cytokine TGF-β within the tumor microenvironment. By inhibiting TGF-β signaling, AdAPT-001 aims to enhance antitumor immune responses and overcome resistance to immune checkpoint inhibitors. The drug is being developed primarily for solid tumors such as soft tissue sarcoma and chondrosarcoma. It can be administered via intratumoral or intraarterial injection[1][3][4][6].
