**ADAPT6-ABD-mcDM1** is an investigational HER2-targeted protein-drug conjugate developed in an academic research setting for HER2-overexpressing cancers. It comprises the engineered HER2-binding ADAPT6 scaffold fused to an albumin-binding domain to extend systemic exposure and conjugated through a maleimidocaproyl linker to the cytotoxic maytansinoid payload mertansine. Following HER2-mediated binding and internalization in tumor cells, the DM1 payload disrupts microtubule function, producing selective cytotoxicity in HER2-high models. It showed antitumor activity in HER2-expressing SKOV3 ovarian cancer xenografts after intravenous administration and remains a preclinical candidate.
