Overview

Adaptive autologous cell immunotherapy is an investigational cell therapy developed by Assistance Publique - Hôpitaux de Paris (AP-HP) for the treatment of metastatic colorectal cancer, specifically targeting patients with unresectable hepatic metastases. The therapy involves the collection of a patient's own circulating lymphocytes via cytapheresis, which then undergo an ex-vivo cell sorting procedure to selectively deplete regulatory T cells (Tregs). Tregs are a subset of T-lymphocytes that typically suppress immune responses and contribute to tumor immune tolerance. By removing these suppressive cells, the therapy aims to enhance the anti-tumor activity of the remaining effector lymphocytes. The treatment protocol includes a lymphodepleting conditioning regimen of cyclophosphamide and fludarabine administered prior to the infusion of the Treg-depleted cell product to facilitate cell expansion and persistence. This approach has been evaluated in Phase I/II clinical trials (e.g., NCT01190163) to assess its safety and efficacy in reducing tumor burden.
