ADAR1-siRNA refers to small interfering RNA (siRNA) designed to sequence-specifically target and knock down the expression of Adenosine Deaminase Acting on RNA 1 (ADAR1). ADAR1 is an enzyme that catalyzes the deamination of adenosine to inosine (A-to-I editing) in double-stranded RNA (dsRNA) substrates. In laboratory and preclinical research settings, ADAR1-siRNA is widely utilized as a gene-silencing tool to investigate the biological roles of ADAR1, particularly its involvement in RNA editing, immune recognition, ribosome biogenesis, and oncogenic translation. Knockdown of ADAR1 using siRNA has been shown to induce type I interferon signaling, trigger apoptosis in various cancer cell lines, and sensitize tumors to therapies such as immune checkpoint blockade or RNA polymerase I inhibitors.
