Overview

**adavosertib + 5-fluorouracil + cisplatin** is a triple combination regimen composed of adavosertib, a selective small molecule inhibitor of Wee1 kinase, 5-fluorouracil, a pyrimidine analog and antimetabolite, and cisplatin, a platinum-based DNA crosslinking agent. - **Adavosertib** inhibits Wee1 kinase, blocking its phosphorylation of cyclin-dependent kinase 1 (CDK1), which leads to loss of G2/M cell cycle arrest, forced mitotic entry, and enhanced cell death, especially in p53-deficient tumors or those with DNA damage[1][5]. - **5-fluorouracil** acts as a thymidylate synthase inhibitor, interfering with DNA synthesis and repair by masquerading as uracil, leading to apoptosis in rapidly dividing cells. - **Cisplatin** crosslinks DNA, prevents replication and transcription, and triggers multiple cell death pathways. Used together, this combination is designed to exploit disrupted cell cycle checkpoints (from Wee1 inhibition) while intensifying DNA damage (from cytotoxics), resulting in synergistic tumor cell killing in cancers such as ovarian, head and neck, and others.
