Overview

Adavosertib (AZD1775) + cytarabine (AraC) is a combination therapy consisting of a WEE1 kinase inhibitor and a nucleoside analog. Adavosertib is a potent and selective ATP-competitive small-molecule inhibitor of WEE1 kinase, while cytarabine is a conventional chemotherapeutic agent that damages DNA. The combination demonstrates synergistic anti-leukemic effects by enhancing DNA damage, abrogating S-phase arrest, and promoting apoptosis rather than cell cycle arrest. This combination shows activity independent of TP53 mutation status, making it potentially valuable for high-risk leukemias with poor prognosis genetic alterations.
