ADB-102 is a novel, non-opioid gene therapy in preclinical development for the treatment of chronic pain associated with knee osteoarthritis. Developed by Adolore BioTherapeutics, it utilizes a replication-defective herpes simplex virus (rdHSV) vector to deliver a modified carbonic anhydrase 8 (CA8*) gene locally to peripheral sensory nerve fibers within the knee joint. The therapy aims to reduce pain by modulating neuronal excitability through activation of Kv7 voltage-gated potassium channels, addressing neuropathic, inflammatory, and nociceptive pain mechanisms. Supported by NIH HEAL grant funding and validated in preclinical models, ADB-102 is being advanced toward IND filing and first-in-human clinical studies[2][3][5][7].
