ADB-103 is an investigational gene therapy being developed by Adolore Biotherapeutics for the treatment of chronic osteoarthritis hip pain. It utilizes the company's proprietary neuromodulation platform, which employs a replication-deficient herpes simplex virus (rdHSV) vector (JDNI8) to deliver the Carbonic Anhydrase 8 (CA8) gene directly to peripheral sensory nerve fibers. The expressed CA8 protein acts as a potent analgesic by modulating intracellular signaling and reducing the excitability of pain-sensing neurons. This localized approach is designed to provide long-term, durable pain relief at the site of the disease while avoiding the systemic side effects and addiction risks associated with traditional analgesics such as opioids.
