Overview

ADB116 is an investigational small molecule drug developed by Jiangsu Aidi Pharmaceutical (also known as Aidea Pharma) for the treatment of acute ischemic stroke (AIS). It functions as a selective antagonist of the P2X7 receptor, an ATP-gated ion channel that is highly expressed in microglia and plays a pivotal role in the neuroinflammatory cascade. By inhibiting the P2X7 receptor, ADB116 prevents the activation of the NLRP3 inflammasome and the subsequent release of pro-inflammatory cytokines such as interleukin-1β (IL-1β) and interleukin-18 (IL-18). This mechanism is intended to mitigate secondary neuronal damage and neuroinflammation following an ischemic event, thereby providing neuroprotective benefits. ADB116 is currently in Phase 1 clinical development, with studies evaluating its safety, tolerability, and pharmacokinetics in healthy volunteers using an oral tablet formulation.
