Drug intelligence / Profile preview
ADC-AVP04 is an investigational antibody-drug conjugate (ADC) developed by Avipep for the treatment of multiple adenocarcinomas, including ovarian, gastric, and prostate cancers. The drug utilizes Avipep's proprietary Avibody™ platform, which consists of a compact, bivalent diabody format derived from the murine monoclonal antibody CC49. ADC-AVP04 specifically targets tumor-associated glycoprotein 72 (TAG-72), a cell-surface glycoprotein highly expressed on various epithelial cancers but minimally expressed in normal tissues. The Avibody is conjugated to the microtubule-disrupting payload monomethyl auristatin E (MMAE) via a trans-cyclooctene (TCO) linker or a protease-cleavable valine-citrulline (vc) linker. In a pre-targeting approach developed in collaboration with TagWorks, ADC-AVP04 is administered first to localize to the tumor, followed by a systemic tetrazine activator that triggers the precise release of MMAE within the tumor microenvironment. This design aims to maximize therapeutic efficacy while minimizing systemic toxicity and avoiding renal clearance.
