ADCE-003

ADCE-003 is an antibody-drug conjugate (ADC) developed by ADCendo ApS, designed to target the urokinase plasminogen activator receptor-associated protein (uPARAP), also known as CD280 or MRC2. The drug consists of a humanized monoclonal antibody directed against uPARAP, conjugated to a potent topoisomerase I inhibitor payload. uPARAP is a collagen-binding endocytic receptor that is highly overexpressed in several high-unmet-need cancers, particularly soft tissue sarcomas (such as osteosarcoma and leiomyosarcoma) and certain epithelial cancers, while showing very limited expression in healthy adult tissues. The receptor's rapid constitutive internalization makes it an ideal target for ADC-mediated delivery. Upon binding and internalization, the topoisomerase I inhibitor is released in the lysosomal compartment, where it inhibits DNA topoisomerase I, leading to DNA strand breaks and apoptosis in the target cancer cells. ADCE-003 is currently in preclinical development for the treatment of soft tissue sarcomas and other uPARAP-expressing malignancies.
