ADCE-D01

ADCE-D01 is a first-in-class antibody-drug conjugate (ADC) developed by Adcendo for the treatment of cancers of mesenchymal origin, with a primary focus on soft tissue sarcoma. The drug consists of an anti-urokinase plasminogen activator receptor-associated protein (uPARAP, also known as Endo180) antibody (AB-004), conjugated to a topoisomerase I inhibitor payload via a protease-cleavable tetra-peptide linker. uPARAP is an endocytic receptor highly overexpressed in multiple subtypes of soft tissue sarcoma and other mesenchymal tumors, but shows low expression in healthy tissues. The mechanism of action involves targeted delivery and internalization into tumor cells expressing uPARAP, followed by release of the cytotoxic payload that inhibits topoisomerase I activity, leading to DNA damage and cell death. Preclinical studies have demonstrated strong anti-tumor activity and favorable toxicology profiles. ADCE-D01 is currently being evaluated as monotherapy in Phase I/II clinical trials for metastatic or unresectable soft tissue sarcoma[1][2][5][6][7].
