AdCh3NSmut is a recombinant, replication-deficient chimpanzee adenovirus serotype 3 (ChAd3) vector-based vaccine candidate developed for the prevention and treatment of Hepatitis C virus (HCV) infection. The vector is engineered to carry a genetic payload encoding the non-structural (NS) proteins NS3, NS4, NS5A, and NS5B of HCV genotype 1b. These proteins are expressed as a single polyprotein that has been mutated (NSmut) to eliminate the enzymatic activities of the NS3 protease and NS5B RNA-dependent RNA polymerase, enhancing safety while maintaining immunogenicity. The vaccine is designed to elicit potent and broad T-cell responses, particularly CD8+ and CD4+ T cells, which are critical for the control and clearance of HCV. It was originally developed by Okairos (acquired by GSK) and is currently associated with ReiThera. AdCh3NSmut has been evaluated in clinical trials as a standalone agent and as part of heterologous prime-boost regimens, often paired with a Modified Vaccinia Ankara (MVA) vector or a human adenovirus serotype 6 (Ad6) vector.
