AdCh3NSmut + MVA-NSmut is a **prime-boost vaccine regimen** for hepatitis C virus (HCV), utilizing two non-replicating recombinant viral vectors. AdCh3NSmut is a modified replication-incompetent chimpanzee adenovirus vector encoding a genetically inactivated NS region (NSmut) of HCV genotype 1b, covering key non-structural proteins (NS3, NS4a, NS4b, NS5a, NS5b). MVA-NSmut uses Modified Vaccinia Ankara (MVA), also encoding the NSmut antigen. The regimen primes the immune system with AdCh3NSmut and boosts the response with MVA-NSmut, inducing strong, broad, and sustained HCV-specific T-cell responses. Both vectors are genetically engineered not to replicate or integrate, with the NS5B polymerase inactivated for safety. Developed for the prevention or treatment of HCV infection, this combination is under clinical investigation and has shown strong immunogenicity and favorable safety in early-phase trials[1][2][3].
