Overview

AdCMVGRPr is a recombinant, replication-deficient adenoviral vector designed for a "genetic radioisotope targeting strategy" in oncology. It contains the cDNA for the murine gastrin-releasing peptide receptor (GRPr) under the transcriptional control of the constitutive Cytomegalovirus (CMV) early promoter. The vector is used to transduce tumor cells, inducing high levels of membrane-bound GRPr expression. Once expressed, these receptors serve as specific docking sites for radiolabeled bombesin (BBN) analogues, such as [125I]-mIP-bombesin or [131I]-mIP-bombesin. This approach aims to overcome limitations of traditional radioimmunotherapy, such as poor tumor penetration and low endogenous antigen expression, by artificially creating a high-density target for radiopharmaceuticals at the tumor site.
