Overview

AdCMVlacZ is a replication-deficient recombinant adenoviral vector, typically derived from Adenovirus serotype 5 (Ad5), that carries the *lacZ* reporter gene under the control of the cytomegalovirus (CMV) immediate-early promoter. The *lacZ* gene encodes the enzyme beta-galactosidase, which serves as a marker to evaluate transduction efficiency and gene expression levels in target tissues through histochemical staining (e.g., X-gal). In clinical and preclinical oncology research, AdCMVlacZ is primarily utilized as a negative control or reporter vector to compare against therapeutic gene therapy candidates, such as those expressing interleukin-12 (IL-12), CD40 ligand (CD40L), or thymidine kinase (tk). It has been extensively used in experimental models of hepatocellular carcinoma (HCC) and head and neck cancers to establish the baseline effects of adenoviral delivery and to validate the specificity of therapeutic outcomes.
