**AdCMVtk** is a nonproprietary, replication-defective recombinant adenoviral gene-transfer vector designating constructs that express the **herpes simplex virus thymidine kinase** suicide gene under control of the cytomegalovirus promoter. Following transduction, HSV thymidine kinase phosphorylates subsequently administered ganciclovir, producing toxic nucleotide metabolites that inhibit DNA synthesis and cause death of transduced tumor cells, with potential bystander killing of neighboring cells. It has been used as an experimental preclinical gene-therapy tool in hepatocellular carcinoma and other tumor models, including in combination studies with adenoviral IL-12 vectors. The label does not define one uniquely standardized commercial product, adenoviral backbone, serotype, or manufacturing lot.
