Overview

ADCT-211 is a novel antibody-drug conjugate (ADC) developed by ADC Therapeutics, targeting the interleukin 13 receptor subunit alpha 2 (IL13RA2), a protein highly expressed in various solid tumors including glioblastoma multiforme (GBM) and melanoma, while having restricted expression in normal tissues. The ADC consists of HuCl47, a humanized IgG1 monoclonal antibody, site-specifically conjugated to the pyrrolobenzodiazepine (PBD) dimer cytotoxin SG2000 via a valine-alanine cleavable linker (PL1801). Utilizing GlycoConnect technology, ADCT-211 achieves a drug-to-antibody ratio (DAR) of approximately 1.8. In preclinical studies, ADCT-211 demonstrated potent and specific anti-tumor activity in IL13RA2-expressing cell lines and xenograft models, including complete tumor eradication in melanoma models and significant activity in glioblastoma models, while maintaining a stable pharmacokinetic profile and favorable tolerability in animal studies.
