ADCT-241

ADCT-241 is a novel antibody-drug conjugate (ADC) developed by ADC Therapeutics. It targets prostate-specific membrane antigen (PSMA), which is highly expressed in prostate cancer cells. The ADC uses an exatecan-based cytotoxic payload, delivered via a proprietary linker technology, to selectively kill PSMA-expressing tumor cells. Preclinical studies have demonstrated that ADCT-241 exhibits potent antitumor activity in both xenograft and patient-derived models of PSMA-expressing prostate cancer and shows synergistic effects when combined with enzalutamide, a standard therapy for advanced prostate cancer. The drug has shown good tolerability in animal models (rats and cynomolgus monkeys). As of 2025, development remains at the preclinical stage[1][2][3][4][5].
