ADCT-701 is an antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody (HuBa-1-3D) targeting delta-like 1 homolog (DLK1), a protein highly expressed during fetal development but restricted in adults except for certain tumors. The antibody is linked via a cathepsin-cleavable linker to SG3199, a pyrrolobenzodiazepine (PBD) dimer cytotoxin. Upon binding to DLK1-expressing tumor cells, the ADC is internalized and releases the PBD payload intracellularly, causing DNA double-strand breaks and apoptosis. ADCT-701 demonstrates both direct cytotoxicity against DLK1-positive cells and bystander killing of neighboring tumor cells. It is being developed primarily for advanced solid tumors with high unmet medical needs including neuroendocrine neoplasms such as neuroblastoma, hepatocellular carcinoma (HCC), small cell lung cancer (SCLC), adrenocortical carcinoma (ACC), and other neuroendocrine carcinomas[1][2][3][5][6][8].
