Overview

AddaVax is a squalene-based oil-in-water emulsion adjuvant that is chemically and functionally equivalent to the MF59 adjuvant used in licensed influenza vaccines. It is primarily used in preclinical research to enhance the immunogenicity of vaccines, particularly for influenza, by promoting a balanced Th1/Th2 immune response and increasing antibody titers. The emulsion consists of squalene droplets (approximately 160 nm in diameter) stabilized by surfactants (Tween 80 and Span 85) in a citrate buffer. In research models, it has been shown to significantly boost hemagglutination inhibition (HI) titers and antigen-specific IgG production. However, its use in certain inflammatory conditions, such as chronic allergic asthma, has been associated with increased eosinophil recruitment at the site of infection following viral challenge.
