ADE513 is an oral, modified-release formulation of tetrabenazine developed by Adeptio Pharmaceuticals for the treatment of tardive dyskinesia. As a reversible inhibitor of the vesicular monoamine transporter 2 (VMAT2), it prevents the uptake of monoamines, particularly dopamine, into synaptic vesicles, leading to a depletion of dopamine levels in the nerve terminals. This reduction in dopaminergic signaling helps to mitigate the hyperkinetic involuntary movements characteristic of tardive dyskinesia. The modified-release technology employed in ADE513 aims to provide a more consistent pharmacokinetic profile compared to immediate-release tetrabenazine, potentially allowing for once-daily administration and reducing the incidence of peak-related adverse effects such as sedation and akathisia.
